چکیده

Carotenoids are purported to provide widespread function in the biology and health of humans and other mammalian species. Provitamin A carotenoids, such as b-carotene, are valued in the diet of many mammals for their contribution as precursors of vitamin A and retinoids. Carotenoids may also function in the prevention of some chronic diseases by improving intercellular communication. enhancing immune response, and operating as antioxidants in vivo. It is widely known that humans and other mammalian species absorb and accumulate carotenoids in body tissues. However, the potential use of carotenoids as modulators of disease and in the prevention of vitamin A de®ciency has been hindered by the limited progress in understanding carotenoid absorption and metabolism. In fact, major gaps in knowledge still exist in the fundamental pathways beginning with release from the food matrix and ending with distribution in body tissues and excretion. Continued development of assessment methods for humans, appropriate animal models for mechanistic studies, and analytical techniques for quanti®cation and identi®cation of compounds is needed to advance our understanding of these critical pathways. This review will discuss the current knowledge involving the fundamental pathways of absorption and metabolism of carotenoids in mammalian species. When applicable, emphasis will be placed on the human.

Mammalian Metabolism of β-Carotene: Gaps in Knowledge

β-carotene is the most abundant provitamin A carotenoid in human diet and tissues. It exerts a number of beneficial functions in mammals, including humans, owing to its ability to generate vitamin A as well as to emerging crucial signaling functions of its metabolites. Even though β-carotene is generally considered a safer form of vitamin A due to its highly regulated intestinal absorption, det...

Intestinal absorption of dietary carotenoids.

The assessment of carotenoid bioavailability has long been hampered by the limited knowledge of their absorption mechanisms. However, recent reports have elucidated important aspects of carotenoid digestion and absorption. Disruption of food matrix and increasing amounts of fat seem to enhance the absorption of carotenes to a larger extent than that of xanthophylls. Comparing different caroteno...

Genetic variations involved in interindividual variability in carotenoid status.

As shown in most clinical studies dedicated to carotenoids, there is a huge interindividual variability in absorption, and blood and tissue responses, of dietary carotenoids. The recent discovery that several proteins are involved in carotenoid metabolism in humans has prompted a possible explanation for this phenomenon: genetic variants in genes encoding for these proteins may affect their exp...

Carotenoid and retinoid metabolism: insights from isotope studies.

Use of isotopes as tracers has had an important role in elucidating key features of vitamin A and retinoid metabolism in animal models and humans. Their use has shown that beta-carotene absorption is variable, and that the appearance of beta-carotene and its metabolites in the blood by time since dosing follows characteristic patterns. Retinol formed from beta-carotene shows a different pattern...

Effects of egg consumption on carotenoid absorption from co-consumed, raw vegetables.

BACKGROUND Dietary lipids are one of the most effective stimulators of carotenoid absorption, but very limited data exist on the impact of endogenous food sources of lipids to enhance carotenoid absorption. The co-consumption of whole egg with carotenoid-rich foods may increase overall carotenoid absorption via lipid-rich egg yolk. OBJECTIVE We designed this study to assess the effects of egg...
